We describe the development of a synthetic strategy to rapidly assembly N-acetylcysteamine (SNAC) thioesters, which are frequently used as substrate surrogates in biosynthesis studies. The strategy bases on cascades of hydroboration and Suzuki-Miyaura reaction with SNAC thioesteracrylates carried out in one-pot. Our report ist the first about a transition metal-catalysed coupling reaction in the presence of SNAC thioesters. These have previously been identified to be particularly problematic due to the sensitivity and dense functionalisation of this functional group.
