Part of Lisa's PhD work was published as part of the Emerging Investigators Series in RSC Advances.
In this proof-of-principle-study we show the first example of a polyketide synthase (PKS) domain immobilisation in form of a cross-linked enzyme aggregate (CLEA). In doing so, we obtained an improved biocatalyst that recovered most of its enzymatic activity, but could be repetitively re-used in semipreparative-scale reactions. Such a cross-linking strategy might be generally helpful to improve the value of PKS enzymes for chemoenzymatic synthesis as these commonly suffer from low stability and activity.
