The results of our collaoration with the group of Prof. Wulf Blankenfeldt at the Helmholtz Centre for INfection Research were published in Angewandte Chemie. We investigated the bifunctional PKS domain AmbDH3 from the amruticin biosynthesis via a combination of structural biology and in vitro biochemistry. Minimal structural deviations and a diagnostic amino acid point mutation were identified, which provide the dehydratase with an additional cyclase activity.
